'In is correct answer for both and not 'on'.
Because as I think so in India now-a-days buses don't allow anybody to sit at the top.

Both "in" and "on" can be used in both the sentences. But I would suggest you to write- (1) I am "in" the car and (2) I am "on" the bus. I'm not sure behind the reasons for doing so. But I think "in" is "generally" {Pay a little attention here. I have written generally.} used for small and personal vehicles whereas "on" is used for public transports. Again, the choice depends on the kind of sentence.

P.S. To tell you the truth, I actually don't know how to explain you this thing!!
