The DNA strand that has not been cleaved is then passed through the single-stranded break. The cleaved strand is then resealed, forming a structure with the same chemical bonds as the starting DNA, but with one less negative supercoil. By this mechanism, the enzyme removes one negative supercoil at a time.

When a twist is unwound without cutting the DNAstrands or is removed by cutting the strand(s) and resealing, negativesupercoils are introduced inthe DNA. But strangely enough unwinding of DNAby helicase cause the DNAahead of the primosome to form positive supercoils.
