The conversion of pyruvate to acetyl-CoA involves the removal of a carboxyl group from pyruvate, resulting in the release of carbon dioxide. This decarboxylation process is catalyzed by a multi-enzyme complex called the pyruvate dehydrogenase complex (PDC). The PDC consists of three enzymes: pyruvate dehydrogenase, dihydrolipoyl transacetylase, and dihydrolipoyl dehydrogenase. The reaction also requires the coenzyme thiamine pyrophosphate (TPP), lipoic acid, and Coenzyme A (CoA).

Production of Acetyl-CoA
During decarboxylation, one molecule of pyruvate produces one molecule of acetyl-CoA. The acetyl group is transferred to CoA, forming acetyl-CoA, while the remaining two-carbon molecule is released as carbon dioxide. Acetyl-CoA is a crucial molecule that enters the Krebs' cycle, also known as the citric acid cycle.
