The foundation of the PCR test is the identification of 16S rRNA molecules from the most prevalent mycoplasma species contaminating cell cultures.
This method's primer specificity should be high enough to detect Acholeplasma as well as other bacteria, but low enough to prevent the amplification of common bacteria that may be present in the PCR reagents.
For the detection of mycoplasma, newly developed techniques like fluorescence in situ hybridization (FISH) or assays based on the detection of adenosine triphosphate (ATP) generation by fluorescence microscopy and luminometer are suggested.
But there is a lack of published data regarding the sensitivity, specificity, and accuracy of either assay when used in routine cell culture.
However, these assays proceed remarkably quickly.
For instance, the findings of a FISH test can be made public in two to three hours, whereas those of a luminescence test can be produced in about twenty minutes.
