  • The shikimate biosynthesis pathway provides precursors for aromatic molecules in bacteria, fungi, apicomplexan, and plants, but not in animals.
  • L-Tyrosine (Tyr) is an aromatic amino acid (AAA) required for protein synthesis in all organisms, but synthesized de novo only in plants and microorganisms.
  • In plants, Tyr also serves as a precursor of numerous specialized metabolites that have diverse physiological roles as electron carriers, antioxidants, attractants, and defense compounds.
  • Some of these Tyr-derived plant natural products are also used in human medicine and nutrition (e.g. morphine and vitamin E).

  • The shikimate pathway consists of seven sequential enzymatic steps and begins with an aldol-type condensation of two phosphorylated active compounds, the phosphoenolpyruvic acid (PEP), from the glycolytic pathway, and the carbohydrate D-erythrose-4-phosphate, from the pentose phosphate cycle, to give 3-deoxy-D-arabino-heptulosonic acid 7-phosphate (DAHP).
  •  The fifth section of the pathway is the activation of shikimic acid with adenosine triphosphate (ATP) (shikimate kinase, SK) to make shikimic acid 3-phosphate (S3P).
  • The sixth chemical reaction is the addition of PEP to S3P to generate 5-enolpyruvylshikimic acid 3-phosphate; the enzyme that catalyzes this reaction step, 5-enolpyruvylshikimate 3-phosphate synthase (EPSPS).
  • Last reaction step of the shikimate pathway is the production of chorismic acid from catalytic action on the chorismate synthase (CS).
  • Further 
    Chorismate mutase catalyzes the rearrangement of chorismic acid to prephenic acid, which is the first committed step in the biosynthesis of aromatic amino acids
  • And prephenate can be converted into 4-hydroxyphenylpyruvic acid; which is catalyzed by the enzyme prephenate dehydrogenase [nadp(+).
  • In yeast, prephenate is involved in the metabolic pathway called the tyrosine metabolism pathway.
