His father, Paigambar Baksh, was a shehnai player in the court of Maharaja Keshav Prasad Singh of Dumraon. Bismillah Khan learned to play the shehnai from his father and he also received training in vocal music from his uncle, Ali Bux 'Vilayatu'.

Bismillah Khan is known for his contributions to Indian classical music. He popularized the shehnai as a solo instrument and he also introduced it to the mainstream music industry. He was a prolific composer and he created several new ragas for the shehnai. He also collaborated with other musicians and he was known for his improvisational skills.

Bismillah Khan received several awards and honors for his contributions to Indian classical music. He was awarded the Padma Shri in 1961, the Padma Bhushan in 1968, and the Padma Vibhushan in 1980.
