Acid hydrolysis is a method used to hydrolyze proteins into amino acids. In the case of A only acid, hydrolysis is carried out and thus it represents the constituent amino acids of the protein. In all other cases, acid hydrolysis is followed by a particular treatment. The carboxypeptidase removes the C-terminal amino acid and in this case, it is an alanine residue, as in B an alanine is separated from all other amino acids. Chymotrypsin cleaves the aromatic amino acids and probably, in this case, it is phenylalanine in C, we can see that there are four amino acids namely: Ala, Lys, Met, and Ser are separated from the phenylalanine set indicating that they come after the phenylalanine. In D, which is treated with trypsin cleavage is occurring between the carboxyl group of arginine or lysine and the amino group of the adjacent amino acid. It gives three fragments indicating that two cleavages had occurred. The first cleavage is between Arg resulting in a small fragment of alanine and arginine and the second is involved between Lys which yields two fragments. In E, the treatment with CNBr is carried out. Cyanogen Bromide or CNBr cleaves methionine residues and in this case, only an Alanine and Serine is separated.
