The Endoplasmic Reticulum (ER) is the main organelle involved in modification and routing of newly synthesized proteins to their destinations.

Explanation:
The Endoplasmic Reticulum (ER) is a network of membrane-bound tubes and sacs that extends throughout the cytoplasm of eukaryotic cells. The ER is classified into two types: the rough endoplasmic reticulum (RER) and the smooth endoplasmic reticulum (SER). The RER is studded with ribosomes, while the SER lacks ribosomes.

The main function of the ER is the synthesis and modification of proteins and lipids. Newly synthesized proteins are translocated into the lumen of the RER, where they undergo post-translational modifications such as folding, glycosylation, and disulfide bond formation. The modified proteins are then transported to their final destinations, such as the plasma membrane, lysosomes, or Golgi apparatus, via vesicular transport.

The ER also plays a crucial role in the quality control of newly synthesized proteins. Misfolded proteins are recognized and retrotranslocated to the cytoplasm for degradation by the proteasome. This process is known as ER-associated degradation (ERAD).

In conclusion, the ER is the main organelle involved in the modification and routing of newly synthesized proteins to their destinations.
