Muscle cells are composed of proteins that work together to enable movement. When muscles contract, the proteins change shape and arrangement, which allows them to shorten and produce force. There are two main proteins involved in muscle contraction:


Actin


  • Actin is a thin, filamentous protein that forms the backbone of muscle fibers.

  • It has binding sites for another protein called myosin.

  • When calcium ions are released in response to a nerve impulse, they bind to a regulatory protein called troponin, which causes tropomyosin to move away from the binding sites on actin.

  • Myosin can then attach to actin and pull it toward the center of the sarcomere, the basic unit of muscle contraction.



Myosin


  • Myosin is a thick, filamentous protein that has globular heads that can bind to actin.

  • When myosin binds to actin, it undergoes a conformational change that allows it to pull actin toward the center of the sarcomere.

  • This movement is called the power stroke.

  • ATP is required to release myosin from actin and allow it to reset for another power stroke.



The interaction between actin and myosin is what causes muscle contraction. When a muscle is at rest, there is a low concentration of calcium ions in the cytoplasm. When a nerve impulse arrives at the muscle fiber, calcium ions are released from the sarcoplasmic reticulum, a specialized type of smooth endoplasmic reticulum, and bind to troponin, causing a conformational change that exposes the binding sites on actin. Myosin then binds to actin and pulls it toward the center of the sarcomere, shortening the muscle fiber and producing force.
