Cannot be used for preparation of vaccines against plague:
1) Avirulent live bacteria:
Using avirulent live bacteria for vaccine preparation against plague is not ideal because live bacteria, even if avirulent, can potentially revert to a virulent form and cause disease in the host. This poses a risk to the individual receiving the vaccine, as well as to others who may come in contact with them. Therefore, live bacteria vaccines are typically not used for diseases like plague where the risk of reversion to virulence is high.
2) Synthetic capsular polysaccharide material:
Synthetic capsular polysaccharide material alone may not be effective for preparing vaccines against plague. While the capsular polysaccharide is an important component of the bacterium that can induce an immune response, it may not be sufficient to confer long-lasting immunity against the disease. Vaccines made solely from synthetic capsular polysaccharide material may not provide the necessary protection against the pathogen.
In contrast, formalin-inactivated suspensions of virulent bacteria and heat-killed suspensions of virulent bacteria are commonly used for vaccine preparation against plague. These methods help to present the antigens of the pathogen to the immune system without the risk of causing disease in the host. By using inactivated forms of the bacteria, the vaccine can stimulate the immune response to produce protective antibodies without inducing illness.
