DNA Polymerase I removes the RNA primer
DNA Polymerase I, also known as Pol I, is an essential enzyme involved in DNA replication in prokaryotes. One of its key functions is to remove the RNA primers that are used to initiate DNA synthesis on the lagging strand during replication.

Process of primer removal
- After the RNA primers have served their purpose in initiating DNA synthesis, DNA Polymerase I comes into play.
- It has 5’ to 3’ exonuclease activity that allows it to degrade the RNA primer by hydrolyzing the phosphodiester bonds between the RNA nucleotides.
- As the RNA primer is degraded, DNA Polymerase I simultaneously synthesizes new DNA in its place using the exposed template strand.
- This process is known as primer removal and is crucial for completing DNA replication on the lagging strand.

Role of DNA Polymerase I in DNA replication
- Apart from removing RNA primers, DNA Polymerase I also has polymerase activity that helps in filling the gaps left behind after primer removal.
- It is involved in both DNA repair and replication processes in prokaryotic cells.
- DNA Polymerase I ensures the accurate and efficient replication of the entire genome by its various activities during DNA replication.
In conclusion, DNA Polymerase I plays a vital role in DNA replication in prokaryotes by removing RNA primers and filling the resulting gaps with DNA. Its exonuclease and polymerase activities are essential for the completion of DNA synthesis on the lagging strand.
