Recombination of V, D and J Ig gene segments:a)Only occurs in mature B...
Recombination of V, D and J Ig gene segments occurs during B-cell development and is responsible for generating the diversity of immunoglobulins (Igs) in the adaptive immune system. The process of V(D)J recombination is mediated by the enzyme complex called V(D)J recombinase, which consists of the proteins RAG1 and RAG2.

Explanation:

Location of V(D)J recombination

V(D)J recombination occurs in developing B-cells in the bone marrow.

Involvement of V, D, and J Ig gene segments

The Ig genes are composed of variable (V), diversity (D), joining (J), and constant (C) region segments. The V(D)J recombination process involves the rearrangement of the V, D, and J gene segments to form the variable region of the Ig molecule.

Mechanism of V(D)J recombination

The V(D)J recombination process involves the cutting and joining of DNA segments. The process is initiated by the binding of the RAG1/RAG2 complex to the heptamer-spacer-heptamer (HSH) recombination signal sequence (RSS) flanking the V, D, or J gene segments. The RAG complex then cleaves the DNA at the border between the RSS and the coding segment, generating a hairpin loop on the coding end. The hairpin loop is opened by the Artemis endonuclease, and the RAG complex mediates the joining of the two coding segments.

Timing of V(D)J recombination

V(D)J recombination occurs during early B-cell development, before the mRNA stage. Once the Ig gene segments have been rearranged, the B-cell will produce a unique Ig molecule with a specific antigen-binding site.

Conclusion:

In summary, V(D)J recombination is a process that occurs in developing B-cells and is responsible for generating the diverse repertoire of Ig molecules in the adaptive immune system. The process involves the rearrangement of V, D, and J gene segments and is mediated by the RAG1/RAG2 complex. V(D)J recombination occurs before the mRNA stage, and once completed, the B-cell will produce a unique Ig molecule with a specific antigen-binding site.
