The correct sequence of steps followed during the process of gene therapy while treating a patient of SCID is as follows:

(i) Retrovirus infects lymphocytes extracted from bone marrow of the patient and cultured.
(ii) Engineered cells are injected into the patient's bone marrow.
(iii) Normal allele is inserted into a retrovirus.
(iv) Retrovirus makes a DNA copy of its RNA. This DNA carrying the normal allele gets inserted into the chromosome of the host cell.

Let's understand the process of gene therapy for SCID (Severe Combined Immunodeficiency) in detail:

1. Retrovirus infects lymphocytes extracted from bone marrow of the patient and cultured:
- Lymphocytes, a type of white blood cells, are extracted from the bone marrow of the patient.
- The extracted lymphocytes are cultured in a laboratory setting.
- A retrovirus is used as a vector to deliver the normal allele (correct gene) into the patient's cells. This retrovirus is modified to carry the desired gene.

2. Normal allele is inserted into a retrovirus:
- The normal allele, which contains the correct gene for the immune system, is isolated and inserted into the retrovirus.
- The retrovirus acts as a carrier or vector for delivering the normal allele into the patient's cells.

3. Retrovirus makes a DNA copy of its RNA. This DNA carrying the normal allele gets inserted into the chromosome of the host cell:
- The retrovirus enters the lymphocytes and releases its RNA.
- The RNA is converted into DNA by the enzyme reverse transcriptase present in the retrovirus.
- This DNA carrying the normal allele is integrated into the chromosome of the host cell (lymphocyte).
- The integration of the normal allele into the host cell's chromosome ensures that the correct gene is expressed and functional.

4. Engineered cells are injected into the patient's bone marrow:
- The genetically modified lymphocytes, now carrying the normal allele, are injected back into the patient's bone marrow.
- These engineered cells have the ability to produce functional immune cells, thereby correcting the immune deficiency in the patient.

In conclusion, the correct sequence of steps in gene therapy for SCID is (iii) normal allele insertion into a retrovirus, (i) retrovirus infects lymphocytes extracted from bone marrow and cultured, (iv) retrovirus makes a DNA copy and inserts it into the chromosome of the host cell, and (ii) engineered cells are injected into the patient's bone marrow.
