Directions: In the following questions, a statement of assertion is followed by a statement of reason. Mark the correct choice as:
Assertion: By insertion of gene encoding complementary RNA only the RNAi can be introduced in an organism.
Reason: In vitro there are no methods by which synthesised complementary RNA can be inserted in an organism to induce RNAi (RNA interference).
  • a)
If both Assertion and Reason are true and Reason is the correct explanation of Assertion.
  • b)
If both Assertion and Reason are true but Reason is not the correct explanation of Assertion.
  • c)
If Assertion is true but Reason is false.
  • d)
If both Assertion and Reason are false.
Correct answer is option 'D'. Can you explain this answer?

RNAi (RNA interference) occurs in all eukaryotic organisms as a method of cellular defense. This method involves silencing of a specific mRNA due to a complementary dsRNA molecule that binds to specific mRNA and prevents translation of the mRNA. To induce RNA interference in any organism in vitro there are two methods, the complementary RNA can be synthesized and then annealed to produce dsRNA and inserted into cells or the cells are genetically modified to produce the two complementary RNA strands or duplex form in vivo by transcription. However, the first method i.e., in vitro insertion of synthesized complementary RNA stops target gene expression for only 4-6 days and their re-introduction is required. But the second method i.e., the vector DNA construct once transfected into the cells, will keep on generating RNA interference and hence, cause a permanent inhibition of the damage.

Understanding the Assertion and Reason
The assertion states that "By insertion of gene encoding complementary RNA only the RNAi can be introduced in an organism." This implies that RNA interference (RNAi) can only be achieved through the introduction of a specific gene that codes for complementary RNA.
Analysis of the Assertion
- RNA interference is a biological process where RNA molecules inhibit gene expression or translation.
- While introducing genes that encode complementary RNA (like small interfering RNA or siRNA) can induce RNAi, it is not the only method. Other techniques, such as direct delivery of synthetic RNA molecules, also effectively induce RNAi.
Understanding the Reason
The reason states, "In vitro there are no methods by which synthesised complementary RNA can be inserted in an organism to induce RNAi." This suggests that it is impossible to use synthesized RNA for RNAi in organisms.
Analysis of the Reason
- This statement is false. There are indeed methods, such as lipofection and electroporation, that allow for the direct introduction of synthetic RNA into cells for RNAi purposes.
- Furthermore, advances in biotechnology have developed various delivery systems, including nanoparticles and viral vectors, for RNA delivery.
Conclusion
Since both the assertion and the reason are incorrect:
- The assertion inaccurately limits the methods to introduce RNAi.
- The reason incorrectly claims the impossibility of using synthesized RNA.
Thus, the correct answer is option 'D': both Assertion and Reason are false.
