The Molecular Basis of Inheritance: Semi-Conservative DNA

- Adenine (A) always pairs with thymine (T) through two hydrogen bonds, while cytosine (C) always pairs with guanine (G) through three hydrogen bonds.

3. Semi-Conservative DNA Replication:
- Semi-conservative DNA replication is a process that occurs during the S phase of the cell cycle.
- The double-stranded DNA unwinds and separates into two individual strands.
- Each separated strand serves as a template for the synthesis of a new complementary strand.
- DNA polymerase enzymes catalyze the addition of nucleotides to the growing DNA strand, following the base-pairing rules.
- As a result, two identical DNA molecules are formed, each consisting of one original (template) strand and one newly synthesized strand.

4. Meselson-Stahl Experiment:
- Meselson and Stahl designed an experiment to determine the mode of DNA replication.
- They grew E. coli bacteria in a medium containing a heavy isotope of nitrogen (15N) for several generations.
- Then, they switched the medium to a lighter isotope (14N) and allowed the bacteria to replicate their DNA.
- The DNA samples were collected at different time points and analyzed using density gradient centrifugation.
- The results showed that after one generation, the DNA had an intermediate density, indicating semi-conservative replication.
